B16-GFP小鼠黑色素瘤細胞-綠色標記 處理步驟被

細胞在培養(yǎng)過程中如果污染了，不要試圖去除污染（非常重要的細胞株才考慮去除污染），一般的原則是應將污染的細胞棄去。
1)細菌污染的排除：可使用大劑量抗生素沖擊法，向污染的細胞培養(yǎng)液中加入大劑量的抗革蘭氏陽性和陰性細菌的抗生素，一般采用5～10倍于常用量進行用藥，加入高濃度抗生素后作用24-48小時，再換入常規(guī)的培養(yǎng)液，有時可以奏效。
2)支原體的排除：卡那霉素、慶大霉素、支原體去除因子(MRA)、卷須霉素及BM-Cycline等對抗支原體是有活性的，可使用它們抑制或消除支原體。

【Complete Growth Medium*培養(yǎng)基】 The base medium for this cell line is DMEM/RPMI-1640 Medium. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%. 
【Subculturing傳代方法】 
Volumes are given for a 75 cm2 flask. Increase or decrease the amount of dissociation medium needed proportionally for culture vessels of other sizes.
Remove and discard culture medium.
Briefly rinse the cell layer with 0.25% (w/v) Trypsin- 0.53 mM EDTA solution to remove all traces of serum that contains trypsin inhibitor.
Add 2.0 to 3.0 mL of Trypsin-EDTA solution to flask and observe cells under an inverted microscope until cell layer is dispersed (usually within 5 to 15 minutes).
Note: To avoid clumping do not agitate the cells by hitting or shaking the flask while waiting for the cells to detach. Cells that are difficult to detach may be placed at 37°C to facilitate dispersal.
Add 6.0 to 8.0 mL of complete growth medium and aspirate cells by gently pipetting.
Add appropriate aliquots of the cell suspension to new culture vessels.
Incubate cultures at 37°C.
Subc*tion Ratio: A subc*tion ratio of 1:2 to 1:6 is recommended
Medium Renewal: 2 to 3 times per week
